A number of monocyte-derived factors that act as potential autocrine modulators of human monocyte functions were isolated and characterized. Human recombinant granulocyte-macrophage colony-stimulating factor (GM-CSF) was employed as a stimulus for human peripheral blood monocytes and three monocyte-derived factors were isolated and characterized: a 31 kDa growth factor (GF31), a 12 kDa GF12 and a 9 kDa GF9. The synthesis of the 31 kDa GF31 and 12 kDa GF12 was partially dependent upon the addition of GM-CSF to the cultures and the production of the 9 kDa GF9 appeared to be constitutive. Exposure of monocytes to recombinant GM-CSF resulted in the rapid loss of several monocyte characteristics including the synthesis of type 1 colony-stimulating factor, 3-hydroxy-3-methylglutaryl coenzyme A reductase, and nitric oxide synthase. The levels of both GF31 and GF12, however, were found to be induced by GM-CSF. In addition, GM-CSF-treated monocytes synthesized a 40 kDa GF40 that was distinct from previously described IL-1 and TNF. This study represents the first direct demonstration of the production of multiple monocyte-derived factors by human peripheral blood monocytes.Connected to the project is the proposed nine-story, 54,000 square foot addition to the existing structure designed by David Burchard Architect, LLC. It is envisioned that the development will include: 184 new market-rate apartments; two new five-story townhouses (one located adjacent to the existing wing); and four new five-story, mixed-income apartment buildings with a total of 118 units.
